Propargylglycine as A Possible Factor of The Diastolic Function Restoration in Old Rats

ABSTRACT

Aging increases the risk of cardiovascular diseases. The objective of the study was to show the effect of propargylglycine (PPG) on the cardiohemodynamics in old rats. We recorded the left ventricular pressure and volume with visualization of pressure-volume curves during the cardiac cycle using ultraminiature catheter 2F (Millar Instruments, USA) to evaluate systolic and diastolic function in vivo. It has been shown that PPG (11.3 mg/kg) in old rats evokes a decrease in both arterial and end-diastolic stiffness 1.5 and 2.1 times, respectively. In addition, we observed PPG-induced elevation of mitochondrial pools of H2S (by 112%), NO2- (by 162%), and enhanced activity of cNOS (3 times), as well as reduced mitochondrial pools of uric acid as a marker of superoxide (·O2- ) generation and ATP degradation. These results suggest that PPG-induced positive effects are determined by enhanced H2S and NO synthesis and, by contrast, suppressed degradation of ATP and ·O2- generation as a marker of oxidative stress.
