Combined Administration of the White Button Mushroom Agaricus bisporus (Agaricomycetes) and Lactobacillus casei Modulated Immune-Related Gene Expression and Mucosal and Serum Immune Parameters in a Goldfish (Carassius auratus) Model
摘要
The aim of this study was to evaluate the effects of white button mushroom (Agaricus bisporus) powder (MP) and Lactobacillus casei, administered either alone or together, on tnf-alpha messenger RNA levels and mucosal and serum immune parameters in goldfish (Carassius auratus). Goldfish (n = 180) were purchased and randomly allocated into 4 groups repeated in triplicate. An 8-week feeding trial was designed and included various treatments: basal diet (control); diet containing 107 colony-forming units [CFU]·g-1 L. casei (Pro); diet containing 1% A. bisporus powder (MP); and diet containing 107 CFU·g-1 L. casei + 1% A. bisporus powder (MP + Pro). The results revealed that combined administration of MP and Pro significantly (P < 0.05) improved expression of tnf-alpha 1 and tnf-alpha 2 genes, total immunoglobulin (Ig) level in the skin mucus, and protease and lysozyme activities in the goldfish. Similarly, serum lysozyme and serum alternative complement activity (ACH50) in goldfish fed MP + Pro was remarkably higher than that in the other groups (P < 0.05). With regard to total serum Ig levels, however, the difference was not significant between the goldfish fed MP + Pro and those receiving the other diets (P > 0.05). The highest expression of immune-related genes (tnf-alpha 1 and tnf-alpha 2), total Ig level in skin mucus, protease and lysozyme activity, and serum ACH50 and lysozyme activity were observed in goldfish fed MP + Pro. These findings reveal that A. bisporus with L. casei can be considered as a promising immunostimulant when administered in the diet in goldfish culture.
