Improvement of Antibacterial Activity of Red Belt Conk Medicinal Mushroom, Fomitopsis pinicola BCC58 (Agaricomycetes), in Fermentation of Lignocellulosic Materials

SINOPSIS

In this study, the antibacterial activity (ABA) of Fomitopsis pinicola BCC58 was evaluated depending on different cultivation conditions. Xylose and glucose followed by cellobiose ensured the highest inhibition of Staphylococcus aureus (74%-97%) and Escherichia coli (51%-80%) growth. Among lignocellulosic materials tested during submerged and solid-state fermentation, the strongest inhibition activity was revealed after fungus growth in the presence of mandarin squeeze. Mandarin squeeze represents an excellent growth substrate for mushroom, increasing the growth inhibition of E. coli and S. aureus by 90-91%, but supplementation of KNO3 or yeast extract favored the increase of ABA. After cultivation in a 7-L fermenter, ethanolic extracts from F. pinicola BCC58 biomass and culture liquid showed the highest ABA, especially against E. coli, with a minimum inhibitory concentration (MIC) of 0.5 mg/mL. Hot water extract from biomass and ethyl acetate extract from culture liquid showed the lowest ABA against E. coli and S. aureus, with an MIC of 15 and 20 mg/mL and 7.5 and 6.0 mg/mL, respectively.
