P.Karst. It was judged homogenous by SDS-polyacrylamide gel electrophoresis. The purification procedure included ammonium sulfate precipitation, ethanol fractionation, DEAE-Sephadex anion-exchange chromatography, crystallization, and recrystallization. Flammulin was crystallized with microdialysis against a polyethylene glycol sodium phosphate buffer. The crystals were rhombohedron-shaped and easy to melt when the crystallization conditions were changed. With a molecular mass of 23,700, flammulin was characterized by its lack of Met. Amino acid composition analysis showed that the numbers of acidic amino acids are more than that of basic amino acids, but the isoelectric point detected by analytical isoelectric focusing was at pH8.9. %8 1999-03-01
