Early Diagnostics of Myocardial Ischemic Injury in Humans Using a Marker of Mitochondrial Permeability Transition Pore Opening

ABSTRACT

The level of mitochondrial factor (MF) as a marker for early diagnostics of ischemic injuries of the myocardium was determined in patients at M.M. Amosov Institute of Cardiovascular Surgery. Reperfusion injuries of the human heart were manifested by mitochondrial permeability transition pore (MPTP) opening and MF release. The MF level remained practically unchanged at the beginning of reperfusion, but 5 minutes after restoration of the blood flow and cardiac contractions it increased to the maximum values. At that, it closely correlated with the data of biochemical studies, which are traditionally used in clinic for diagnostics of the heart damages. This enables reperfusion to be a major damaging factor to the myocardium, involving peripheral vascular bed to maintain cardiac muscle viability. It is also important that, a change in MF level was marked just at first minutes of heart reperfusion, which is much earlier than that revealed using other markers. So, investigation of MF level seems to be very perspective for early diagnostics of ischemic damages to different tissues and organs.
