Cryopreservation of wild isolates of Nostoc linckia (Roth) Born. et Flah.

We present a simple method to preserve cultures of cyanobacterium Nostoc linckia (Roth) Born. et Flah. during experiments (5-8 weeks), emphasizing easy-handled preservation of mucilaginous thallus, including spores. The isolates were collected from various parts of Mount Carmel National Park, Haifa, Israel. Collections were made specifically from extreme environments, such as dry, moist, dark, shaded places etc. We tried two methods to preserve cultures: one with liquid paraffin at two different temperatures (-20 °C and 4 °C), and another with dried cultures. In all cases percentage of revival was excellent. Preservation of cultures of N. linckia in liquid paraffin at -20 °C was better in comparison to preservation of those cultures at 4 °C. Revival of the dried cultures took longer than those preserved with liquid paraffin. We also found that the long revival period for dried cultures can lead to bacterial contamination.
