Pioglitazone, an Activator of Peroxisome Proliferator-Activated Receptor (PPAR-γ), Reduces the Expression of KB Nuclear Transcription Factor and Inhibits Apoptosis in Peripheral Blood Mononuclear Cells in Vitro

To examine the impact of PPAR-y activator on the expression of kB nuclear transcription factor (NF-kB) and apoptosis, the peripheral blood mononuclear cells (PBMC) were incubated in the presence of pioglitazone at a final concentration of 10, 30, and 100mmol/l. An expression of NF-kB in CD64+-cells and apoptosis were determined by flow cytofluorometry using appropriate monoclonal antibodies. The development of apoptosis was evaluated by using annexin V and propidium iodide. Activation of PPAR-γ led to a reduction of both a number of CD64+-cells and maximum levels of the receptor expression, as well as a dose-dependent decrease in the NF-kB concentration in the PBMC suspension and CD64+-cells. Analyzing apoptosis, we observed a reduction of apoptotic cells (AnV+ PI+-cells). The obtained results indicate that PPAR-γ activation is involved in the regulation of apoptosis in the PBMC and expression of NF-kB transcription factor, testifying that PPAR-γ is involved in the control of NF-kB activity, playing a leading role in the realization of inflammation, oncogenesis, and so on.
