Analysis of Tumor Necrosis Factor-Alpha (TNF-α) mRNA and Its Receptors in Single Murine Oocytes during Oocyte Meiotic Maturation using Reverse Transcription Polymerase Chain Reaction (RT-PCR)

The study was conducted to investigate the expression of tumor necrosis factor-α (TNF-α) and its receptors (types I and II) in murine oocytes at germinal vesicle and the first polar body stages. Oocytes with intact germinal vesicles were isolated from the mouse ovaries and subjected to in vitro maturation to obtain oocytes forming the first polar body. A reverse transcription polymerase chain reaction (RT-PCR) was used to examine the expression of TNF-α and its receptors at mRNA level. It has been shown that mRNA TNF-α was expressed in single oocytes, and its level was decreasing in the course of transition from germinal vesicle to the first polar body stage. At the same time, TNF-receptor expression was not observed in the single oocytes. Thus, these data are an important link to elucidate the molecular mechanisms regulating oocyte maturation, as well as follicle development.
