Antitumor Effect of Combination Therapy of Proflamin with Cryosurgery of Meth-A Fibrosarcoma in BALB/c Mice

Proflamin (PRF), an antitumor agent, is orally effective against the syngeneic tumors in mice and is a new biological response modifier, which was isolated from the culture mycelium of Flammulina velutipes (W. Curt.: Fr.) Singer. Cryosurgery was used against tumors in this study in which cryoimmune effects were revealed. In this study, immunomodulation with PRF administration was evaluated in combination with cryosurgery. Cryosurgery was carried out on Meth-A fibrosarcoma 7 days after the inoculation of tumor cells into BALB/c mice. The tumor-bearing mice were found to have acquired tumor immunocompetence against the Meth-A tumors after receiving cryosurgery. This tumor immunocompetence reached nadir 2 weeks after the cryosurgery and increased gradually thereafter, reaching its maximum level at 5 weeks and remaining there for 7 weeks. Therapy in combination with cryosurgery and the immunopotentiator PRF was found to abolish the depression of tumor immunocompetence 2 weeks after cryosurgery. PRF was found to augment tumor immunity caused after cryosurgery, and thus it is coincident with modulation. The combination of cryosurgery and an immunopotentiator is suggested to be effective for maintaining the immunocompetence at a high level for a long duration and may be a useful tool in treating cancer.
