Effect of Ukrain on Cell Survival and Apoptosis in the Androgen-Independent Prostate Cancer Cell Line PC-3

Prostate cancer is the most common malignancy and second leading cause of cancer death in male population in all developing countries. Ukrain is a reaction product of different alkaloids from Chelidonium majus L. (celandine) conjugated with thiophosphoric acid. It has cytotoxic effects on various malignant cells. Cell viability was assessed by MTT method in PC-3 cells after the treatment of Ukrain. The IC50 value was observed in 10 μg concentration of Ukrain. Bax, Bad and FasL mRNA expressions were analysed by RT-PCR and protein expressions of p- Akt, Bcl-2 and caspase 10 were analysed by western blot method. Nuclear staining was detected by DAPI staining. Ukrain significantly increased the proapoptotic mRNA expressions of Bad, Bax and FasL. Ukrain decreased the cell survival protein p-Akt and also antiapoptotic protein Bcl-2. Ukrain increased the protein levels of cleaved PARP and caspases-10. This study suggests that Ukrain decreases the cell survival of androgen independent prostate cancer cells and induces apoptosis, which highlights the importance of Ukrain as a therapeutic drug for the androgen independent prostate cancer treatment.
