Macrophage Stimulation by Polysaccharides Isolated from Barometer Earthstar Mushroom, Astraeus hygrometricus (Pers.) Morgan (Gasteromycetideae)

ABSTRACT

The present study investigates macrophage-stimulating polysaccharides isolated from the Barometer Earthstar gastroid mushroom, Astraeus hygrometricus. The alkaline-extracted and ion exchange−purified polysaccharide was designated AE2. This polysaccharide is composed of 69% glucose, with a high molecular weight. It was studied with respect to macrophage-stimulating activity in vitro and exhibited enhanced production of nitric oxide and cytokines. The phagocytosis by AE2-treated macrophages was more than twofold that of the controls. In addition, there was a higher surface localization of lipid rafts upon treatment. The mode of action of AE2 on macrophages may be attributed to its binding to the cell surface, followed by its internalization.
