Ecdysterone Prevents Lowering of Erythrocyte Resistance to Acid Hemolysis in Focal Cerebral Ischemia-Reperfusion

RÉSUMÉ

We studied erythrocyte resistance to acid hemolysis in cerebral venous outflow of adult rats in dynamics of focal ischemia (on the 15th, 30th, 45th, and 60th min), as well as in early (5 min) and remote (24 hours) reperfusion periods. Those rats were treated prophylacti-cally with ecdysterone (1 mg / kg) in the form of a standardized extract of the herb Serratula coronata for 18 days, per os. The analysis of kinetic curves of acid hemolysis showed a considerable increase (almost 60 times, from 1.45 to 85.85% in 60 min of ischemia and 5 min of reperfusion, respectively) in the fraction of erythrocytes with low resistance to acid hemolysis in control rats not receiving ecdysterone. In erythrocytes protected with ecdysterone, such significant changes did not occur, indicating a considerable increase in erythrocyte resistance to acid hemolysis evoked by the prophylactic introduction of the drug. Significant reduction of elevated, especially in the early period of reperfusion, pools of diene conjugates and leukot-riene C4 indicates the possible antiradical mechanism of ecdysterone-induced protective action.
