Flutamide-Induced Blockade of Androgen Receptors Affects the Liver External Secretory Function in Rats

The effects of flutamide (25 mg/kg, intragastrically, daily for three days) on bile formation in male rats were investigated in in vivo experiments, and the bile acid composition of bile was analyzed. The volume rate of bile secretion decreased in male rats under the influence of flutamide, as well as the concentration of glycoconjugated bile acids in the bile of male rats under flutamide action. In particular, biliary glycochenodeoxycholate and glycodeoxycholate concentrations are significantly lower (by 16.3-34.5%) in male rats under flutamide action, compared to those in bile samples of the control group of animals. In this situation, flutamide fails to change cholate conjugation factor calculated as bile acids/unconjugated bile acids ratio. Flutamide inhibits hydroxylation of bile acids in male rats. Coefficient of bile acids hydroxylation is lower under the influence of flutamide, compared to the control value by 10.1%. Thus, flutamide significantly decreases bile flow, bile acids conjugation with glycine, and glycoconjugate transport into bile.
