Contribution of Eicosanoids to the Mechanisms of Contractile Responses of the Portal Vein to Adrenaline and Noradrenaline

The effect of indometacine (7·10−8 M) on the adrenaline (1·10−5 M) − and noradrenaline (5·10−6 M)-induced tonic contractile activity of the isolated rat portal vein (PV) was studied. Indometacine decreased the amplitude of noradrenaline-induced tonic constriction of the PV with intact endothelium, and it produced no effect on constriction of those vessels with denuded endothelial layer. The results obtained suggest that these responses are mediated by endothelial prostanoids. An inhibition of adrenaline-induced constriction of the PV preparations with intact and denuded endothelium indicates that the contractile responses are mediated by prostanoides, derived from both endothelial and smooth muscle cells.
