Effect of Sodium Hydrosulfide (NaHS) on Oxidative/Nitrosative Stress and Endothelium-Dependent Relaxation in Old Rats

The effects of hydrogen sulfide donor, sodium hydrosulfide (NaHS) on indicators of oxidative and nitrosative stress, coupling of constitutive NO-synthase (cNOS) and endothelium-dependent relaxation of smooth muslles (SM) were studied in old rats. In the aorta of old rats, a combined oxidative and nitrosative stress developed resulting in cNOS uncoupling and decreased constitutive synthesis of NO. The biochemical changes correlated with lowering of the endothelium-dependent relaxation of aortic smooth muscles (7.5 ± 1.4%, compared with 64.9 ± 3.5% in control group of adult animals). It has been shown that NaHS significantly inhibited oxidative /nitrisative stress, restoring cNOS coupling and increasing constitutive synthesis of NO. The latter promotes an improvement of endothelium-dependent vasorelaxation by increasing (by 6.5 times) Ach-induced relaxation of aortic smooth muscles.
