Effects of Stimulation and Blockade of Endogenous Hydrogen Sulfide Synthesis in Myocardial Ischemia-Reperfusion

In experiments on isolated rat hearts, perfused according to the Langendorf method, the effects of stimulation and blockade of endogenous hydrogen sulfide synthesis during myocardial ischemia-reperfusion (20 min / 40 min) were studied. A precursor of endogenous hydrogen sulfide synthesis, L-cysteine (121 mg / kg, i.p), was administered 30 min before the start of the experiment. In a similar way, the latter was introduced in 10 min after administration of DL-propargylglycine (11.3 mg/kg; "Sigma", USA), an inhibitor for cystathionine- γ-lyase, an enzyme that synthesizes hydrogen sulfide from L-cysteine. The degree of disorders in mitochondrial membrane permeability, leading to the formation of mitochondrial permeability transition pores, was assessed from the release of a stable mitochondrial factor (λ = 250 nm) in the coronary outflow probes during the initial phase of reperfusion. It has been shown that stimulation of endogenous hydrogen sulfide synthesis by introduced L-cysteine resulted in a decrease in reperfusion dysfunction of the heart, compared to control rats. The introduction of L-cysteine, on the background of inhibited by DL-propargylglycine cystathionine-γ-lyase, rendered a powerful cardioprotective effect in ischemia-reperfusion injury, which was manifested in a significant reduction of reperfusion disorders of cardiodynamics and the contractile function of the myocardium, and improved efficiency of the oxygen metabolism. Positive effect of the combined action of DL--propargylglycine and L-cysteine was also realized through prevention of mitochondrial permeability transition pore opening.
